Would two smaller diffusers at each side of the tank help provide better co2 distribution than 1 large diffuser at one side?

it depends. You need to position the diffuser(s) in the path of water flow where the bubbles get blasted around the tank, if you can do this with 2 diffusers then go for it, but if the bubbles will be rising to the top then there isnt much point to it.

If you can position the diffuser below the outlet of the filter or powerhead/pump then a single diffusers can work fine. I've got mine in my 4ft going into my 3000lph powerhead which sends the bubbles all over the place. Also if you use two diffusers you've got to find a way of splitting the feed tube so it can work on two diffusers, why make work for yourself? :)
